Mechanical unfolding of a guanine (G)-quadruplex! A bimolecular G-quadruplex was formed between a pair of single-stranded DNAs on an AFM probe and a flat gold surface (see figure). The resulting G-quadruplex was studied by single-molecule force spectroscopy as a function of K+ concentration and the force loading rate. The mechanical stability and kinetic and thermodynamic parameters of the G-quadruplexes were estimated.
