The invention discloses a preparation method of 4-amino-3-quinoline formonitrile derivative. The preparation method comprises the following steps of: condensing and cyclizing 3,4-disubstituted aniline (IIa) and 3,3- dimethoxy-2-formyl propionitrile (IIb) to generate 6,7-disubstituted-3-quinoline formonitrile (III), oxidizing and nitrifying an intermediate (III) to generate 6,7-disubstituted-4-nitryl-3-quinoline formonitrile-N-oxide (IV), and the intermediate (IV) is reduced to generate the 4-amino-3-quinoline formonitrile derivative (I). The 4-amino-3-quinoline formonitrile derivative is an important intermediate for preparation of an anti-tumor medicine with 3-nitrile quinoline parent nucleus structure, and for the preparation method of the 4-amino-3-quinoline formonitrile derivative, raw materials are easy to get, the technology is compact, and the method is economical and environmental-friendly, and is suitable for industrial production.
